What is Nitric Oxide?

•    Nitric Oxide or NO, a highly unstable gas, occurs when Nitrogen (symbol N) and Oxygen (symbol O) join together.

What Isn’t Nitric Oxide (NO)?

•    Nitrous Oxide (laughing gas) is the chemical compound N2O. It is a colorless, almost odorless gas, first discovered in 1793 by the English scientist Joseph Priestley. It is a much more stable gas than NO.

•    Nitrogen dioxide (NO2), as a reddish-brown gas with a characteristic sharp, biting odor. NO2 is one of the most prominent air pollutants and a poison by inhalation.

NO vs ADNO

•    NO therapy – medicine often sees NO therapy as the administration of actual Nitric Oxide gas (for example: as administered to premature infants with undeveloped lungs).

•    As NO can convert to poisonous NO2 when exposed to oxygen, direct NO can be toxic and deadly.

ADNO therapy

•    Arginine Derived Nitric Oxide uses Nitric Oxide pre-cursers to allow the body to produce it’s own Nitric Oxide via Nitric Oxide Synthases which convert Arginine to Nitric Oxide. 

Nitric Oxide Synthases

•    At this time three distinct Nitric Oxide Synthases (NOS) have been discovered falling into two categories:

•    1) constitutive – endothelial and neuronal NOS. These synthases are calcium dependent and can be drive by hormones in the body such as estrogen and progesterone (i.e. pregnancy).

•    2) inducible – inducible NOS. This synthase is not calcium dependent and is driven when the immune system is attacked such as in the case of an infection or injury.

Nitric Oxide: Pioneering PC treatment?

Researchers from the University of Utah and the National Cancer Institute developed an experimental drug that reacts with a substance inside cancer cells, releasing nitric oxide to kill the cells or slow their growth without harming healthy cells.

Tests showed the drug, called JS-K, induced the cell-suicide process in acute myeloid leukemia (AML) cells grown in culture and inhibited their growth in mice. In other tests on cell cultures, the drug also inhibited growth of AML cells and - to a lesser extent - prostate, colon and breast cancer cells. It also inhibited growth of prostate cancer cells in mice.

"If all goes well, this would be the first cancer chemotherapeutic agent based on selective delivery of nitric oxide to tumor cells and therefore would constitute a novel class of cancer drugs. It looks promising in cell culture and mice, but much work needs to be done before we know if it can be used in humans for the treatment of cancer.“ - Paul Shami, MD, associate professor of medical oncology at the University of Utah School of Medicine.
